Reducing the size of a large file

After working on files in Photoshop, with multiple masks, they get to be very large- 300-500MB. They are saved as tiffs. I am working off a copy of the original image. I need to reduce the size to submit for publication. Can I flatten the file to reduce the size? Will flattening have any adverse effect on the image? Is there a general rule when an image should be flattened? Thanks for any help.

Yes, you can and should flatten it before submitting. Flattening will not have adverse effects on the image other than not allowing someone to edit your adjustments.

If the publication requires a specific physical size and/or file size you may also need to resize.
