Scott, there's only one Grand Teton (no "s"). Moose usually hang out in willow groves and meadows. There's some good groves around Oxbow Bend where they often hang out.
Not sure what you mean by "how to shoot" - same as any other place. Sunrise is a great time to be out along the main road - the mountains light up at sunrise. The classic landscape views are from Schwabacher's Landing, Snake River Overlook and Oxbow Bend - all a short distance from the main road. Another very common view is of the Mormon Row barns, also at sunrise. You can get there from the Antelope Flats road.

In Yellowstone the Lamar Valley is usually productive for Moose and Bear and the occasional far away view of Wolf. I am not sure about the Tetons. You say you are going for a weekend? I think a week is a minimum to give yourself the best opportunities - especially since Grand Teton is an hour drive from Yellowstone's center.

I have usually encountered moose in the stretch of river on either side of the boundary between Yellowstone and Grand Teton. I can't remember now which side of the boundary, but it's a lowland area with the road fairly close to the river. They are usually so visible here however, that they attract quite a crowd when they appear. In Yellowstone itself I can almost always find them upstream a mile or two on Indian Creek. Haven't been there in a few years, but it was reliable in the past. As Guy says, mornings are usually best.

Near Jackson Lake Lodge can be productive this time of the year. There is a sow grizzly with 3 cubs in the area between there and the ox bow so be carefull where you walk.
Make noise

The best place last year was the Sawmill Ponds area along the Wilson-Moose Road. We were able to get sown in the willow thickets with three moose - a bull, a cow, and last year's calf. There was one addititional moose in the near area. For a couple years running, there were moose hanging out in and near the Gros Ventre Campground - you might check with the campground people about moose sightings near there. We also have seen moose along the Snake River near the Moose Visitors' Center (as if moose can read!!), as well as in the Schwabachers' Landing area. Last year there was an enormous bull moose in the willow thickets just North of the Jackson Lake dam; quite visible from the road. As already mentioned, the Oxbow area is also good for moose sightings. And, as also mentioned, sunrise and twilight are excellent times to watch for moose, as they are active during cooler times. During the heat of the day they tend to be in sheltered areas out of sight.

I've shot moose at sunrise at Schwabacher's Landing, and in the evening at Oxbow Bend. I've also seen them often along the main highway near Moran Junction, and in a patch of trees southwest of Gros Ventre Junction (there's a pull-off from the main highway).

Buffalo should also be pretty easy to find. They tend to hang around the Antelope Flats area. I've also seen them along the road up near Moran Junction.

Another good spot I've found for wildlife is the Kelly Warm Springs. Early in the morning, this is a good spot to find trumpeter swans and other wildlife hanging out in the warm water. I've only been here in fall and winter, though, so I don't know how active it is in spring and summer.

Grand Teton has more Moose possibilities than Yellowstone. The best place for Moose in GT is the Oxbow Bend/Willow Flats/Jackson Lake Lodge area. Also, there are some ponds and willows near the Moose end of the Moose-Wilson Road that can be good for Moose.

In Yellowstone, Moose are most often seen in the Lost Lake/Petrified Tree area. Currently there is a cow Moose with two calfs hanging around the Floating Island area. The other place in Yellowstone for Moose is the Lewis River downstream from Lewis Falls.
